概要 The metastasis via lymphatic system is known to be the first step to distant metastasis, however, little is known about the molecular mechanisms of cancer cell interaction to the lymphatics. Previously we reported that the migration of pancreatic cancer cell BxPC-3 was suppressed by reactive oxygen species (ROS), at the 77th Annual Meeting. Here, we analyzed the role of ROS generating enzyme NOX4 on human dermal lymphatic endothelial cells (HDLEC) in BxPC-3 migration.
Firstly, we evaluated the migration of BxPC-3 cells with or without HDLEC and found that BxPC-3 cells migrated naturally towards HDLEC without any stimulation. Next, we knocked-down NOX4 protein expression on HDLEC by siRNA and evaluated the migration of BxPC-3 cell towards HDLEC. We found that the migration of BxPC-3 cells towards NOX4-knocked-down HDLEC was enhanced, particularly in the velocity.
These data suggest that NOX4 on LEC plays an important role on the cancer cell migration towards lymphatics. ROS in cancer micro-environment may have a negative effect on cancer cell migration.
