概要 We detected superoxide anion generated by human granulocytes using lucigenin-enhanced chemiluminescence (LECL). About 80% LECL activity remained even after complete removal of cell suspension ten minutes after PMA stimulation. LECL by leukocyte adhesion deficiency (LAD) granulocytes disappeared when cell suspension was removed. And the LECL by LAD granulocytes was one-fifth of that by normal granulocytes. LAD granulocytes produced superoxide anion to normal extent detected by SOD-sensitive cytochrome c reduction or Oxygen consumption. About 20 % of normal granulocytes were adhered to reaction tube ten minutes after PMA stimulation whether cell suspension contained lucigenin or not. These results indicate that LECL by a adhered granulocyte is extremely higher than that by a suspended one. There was no difference of superoxide production between adhered granulocytes and suspended ones, when it was detected by luminol-enhanced CL or CLA-enhanced CL. These results indicate that superoxide generated by granulocytes is not enhanced by adhesion. So we conclude that adhered granulocytes have a unique mechanism to elevate LECL.
