概要 | Heat shock protein(Hsp90) has numerous cellular functions, including promotion of proper protein folding, and stabilizes a number of oncogenic proteins, many of which are overexpressed in cancers. The objective of this study is to evaluate an antitumor effect of Hsp90 inhibitor, called 17-AAG, in neuroendocrine tumor (NET) cell lines. NET cell line (COLO320) was treated with Hsp90 inhibitor, 17-AAG, to examine the effect on proliferation of NET cells by using the WST-1 proliferation assay.
17-AAG inhibited cell proliferation in NET cells in a dose- and time-dependent manner. 17-AAG was also effective to induce apoptosis in NET cells as determined with FACS analysis and immunochemistry. The Hsp90 inhibitor is useful as an antitumor drug in NET, inhibiting the proliferation of NET cells through novel mechanism. |
