概要 Presently, more than 20 different keratin (cytokeratin) intermediate filament proteins have been identified, but their biological roles remain largely unknown. In this study, the distribution of 11 keratins (keratin-3, 5, 7, 8, 10, 13, 14, 17, 18 and 20) and actin was studied immunohistochemically and ultraimmunohistochemically using an avidin-biotin peroxidase complex system. Epithelia of both interlobular and intralobular ducts consisted of columnar and basal cells. In the columnar epithelial cells, the intermediate filaments were concentrated near the apex region and formed a thick network, apex network. The upper layer of the apex network contained keratins 7, 18 and 20. In contrast, the lower layer of the apex network contained keratins 3, 5, 8, 17, 18 and 20. Since the filaments of the apex network are tightly anchored to desmosomes, these keratins serve to maintain cell-cell contact. In addition, intermediate filaments containing keratin 7 were anchored to semidesmosomes which served to maintain cell-matrix contact. Keratin 8, keratin 14 and actin were collocated near the cell nucleus and formed a complex cage (about 1μm in diameter) surrounding the Golgi apparatus. The cage may be involved in the positioning and maintenance of specific structure of the Golgi apparatus. In the basal cells, the intermediate filaments concentrated in the perinuclear region and form a juxtanuclear network. This network consisted of keratins 3 and 10. Keratin 13 was detected between the juxtanuclear network and cell membrane. Therefore, keratin 13 may play a role in cell-cell and cell-matrix contact in the basal cells.
